Tumor-promoting Phorbol Esters

چکیده
The effect of tumor-promoting phorbol esters on the in vitro proliferation of mouse pluripotent hematopoietic stem cells (CFU-S) was examined using a short-term in vitro culture system and an 11-d spleen colony assay. Phorbol myristate acetate (PMA, 10-7 M), but not the inert compound phorbol, supported the in vitro survival of day 11 CFU-S for 72 h in a manner similar to IL 3. PMA also enhanced the effect of IL 3 on the in vitro survival of day 11 CFU-S and as little as 1 h of exposure to PMA was sufficient for this purpose. The effect of PMA on CFU-S survival in vitro was not mediated by prostaglandins, did not require an established adherent cell population, and was observed at a concentration of 10M. PMA alone did not enhance the in vitro survival of day 11 CFU-S at very low concentrations of FCS but was still able to potentiate the effect of IL 3 on these cells. PMA also enhanced the in vitro survival of day 11 CFU-S from mice treated with 5-fluorouracil or from marrow cells exposed to merocyanine 540 and light. The interaction of PMA with day 11 CFU-S was not inhibited by a neutralizing antiserum to IL 3 but was inhibited by the protein kinase inhibitor H-7. Together, the data indicate that tumor-promoting phorbol esters interact with pluripotent hematopoietic stem cells. Like IL 3, their effect appears to be permissive and involves stem cells with marrow repopulating ability.

Tumor-promoting phorbol esters stimulate hematopoietic colony formation in vitro.
Tumor-promoting phorbol esters stimulated mouse bone marrow cells to form myeloid colonies in agar cultures without added colony-stimulating factors. The colony-stimulating ability of various phorbol esters correlated well with their ability to promote skin tumors in vivo. These results suggest that phorbol esters mimic the action of specific colony-stimulating factors that regulate growth.

Esters of phorbol, a family of tumor-promoting agents, interact in vitro with cellular membranes. In CsCl equi librium density gradients, isotopically labeled phorbol didecanoate cosediments with microsomal and nuclear membranes prepared from rat liver. The interaction be tween these membranes and the promoting compound is enhanced in the presence of native or denatured DNA. Moreover, changes i...

Phorbol, the terpene alcohol found in the skin tumor-promoting components of croton oil, was isolated, and four diesters were synthesized from it. The tumor-promoting potencies of these compounds were tested in Charles River GDI mice and skin tumor-susceptible mice. The esters of high tumor-promoting ability (12-O-tetradecanoyl-phorbol-13acetate and phorbol-12,13-didecanoate) were effective in ...
متن کاملStimulation of the synthesis of mouse epidermal histones by tumor-promoting agents.
Topical application of 17 nmoles of the potent tumor promoter, 12-O-tetradecanoylphorbol-13-acetate, resulted in a stimulation of the incorporation of [(3)H]lysine into epidermal histones. Maximum incorporation occurred 24 hr after treatment, concurrent with maximum DNA synthesis. The effects of phorbol and two phorbol esters on histone synthesis were related to their tumor-promoting activities...

DNA-histone interaction resulting in increased gene activation (1 , 13). In an effort to observe any existing correlation between the phosphorylation of mouse epidermal histones, tumor promotion, and the increased epidermal macro molecular biosynthesis caused by phorbol esters, we have studied the effect and specificity of some phorbol esters on histone phosphorylation, both in the intact mouse...
متن کاملInduction of plasminogen activator in cultured cells by macrocyclic plant diterpene esters and other agents related to tumor promotion.
In vitro systems that are responsive to tumor-promoting agents may facilitate the identification of such agents and the analysis of their mode of action. We have previously reported that the potent tumor promoter phorbol-12-myristate-13-acetate induces the synthesis of the enzyme plasminogen activator in cultured chick embryo fibroblasts. We have, therefore, tested various compounds for their a...
